摘 要:目的:采用基因表达谱芯片技术筛选丹红注射液治疗心肌梗死作用相关的差异表达基因,初步探讨丹红注射液治疗缺血性心脏病的作用机制。方法:建立左前支冠状动脉结扎大鼠心肌缺血模型,分组给药后分离提取梗死心肌旁的缺血心肌组织总RNA,与大鼠全基因表达谱芯片杂交后进行检测。对筛选得到的药物治疗过程中的差异表达基因进行生物信息学分析,同时对芯片筛选结果进行实时荧光定量PCR(RT-qPCR)验证。结果:丹红注射液治疗组与模型组相比,共筛选得到171条差异表达基因,主要与蛋白结合、转录因子活性及钙离子结合等功能相关。结论:丹红注射液可能主要通过调节蛋白结合,转录因子释放及钙离子结合达到治疗心肌缺血所造成的心肌损伤及梗死的目的。The study is aimed to screen different gene expressions during myocardial infarction(MI) treated by Dan Hong(DH) injection.The mechanism of DH injection was initially investigated in the treatment of MI.The MI model of rats was created by ligating left anterior coronary.Ischemic heart tissues were collected for future analysis.The total RNA was extracted and used to detect different expression genes by rat cDNA microarray.Differential gene expressions were analyzed by bioinformatics analysis.Result of microarray screening was verificated by real-time PCR method.The result showed that obtained 171 different expression genes,which were primarily related with protein binding,activity of transcription factor,and calcium ion binding.It is concluded that releasing transcription factor,regulating protein binding and calcium binding may be the main mechanism of DH injection against MI.
