摘  要:建立了微生物混合转化井冈霉素制备井冈霉烯胺的方法。研究了SIPI-VA18和SIPI-V08单菌转化及两菌混合转化的特点,考察了接种量、两菌混合比例和转化温度的影响。底物井冈霉素浓度5%,菌体浓度70mg/ml,SIPI-VA18和SIPI-V08的浓度比60:10,30℃转化72h,底物降解率为100%,转化液中的井冈霉烯胺浓度为6.07 mg/ml,摩尔转化率为53%。A mixed microbial conversion method was established for the preparation of valienamine from validamycin. The conversion characteristics of two strains, SIPI-VA18 and SIPI-V08, were studied. The reaction mechanism of the combination of SIPI-VA18 and SIPI-V08 was also found to vary from the two single strains. The effects of the inculation amount, the ratio of two strains and the temprature on the conversion were investigated and the optimized conditions were as follows: validamycin concentration of 5%, inoculums concentration of 70 mg/ml, the ratio of SIPI- VA18 to SIPI-V08 of 60 : 10, conversion at 30 ~C and the cultivation time of 72 h. Under the above conditions, the substrate degradation rate was 100 % and the concentration of valienamine was up to 6.07 mg/ml with molar yield of 53 %.
