摘 要:目的运用祖国传统医学“药食同源”的原理,研究香菇多糖(Lentinan,LTN)对实验性高血糖小鼠血糖、糖耐量、肝糖原和血清胰岛素水平的影响,旨在开辟治疗糖尿病的新途径。方法体重(20±2)g清洁级BALB/C小鼠30只腹腔注射四氧嘧啶(120 mg/kg)建立糖尿病动物模型,并将其随机分成4组,分别用生理盐水(A)、低剂量(125 mg/kg)(B)和高剂量(250 mg/kg)香菇多糖(C)以及盐酸二甲双胍(D)灌胃,试验期21 d,观察、比较香菇多糖的作用。结果香菇多糖有明显的降血糖作用,低、高剂量组的血糖水平由实验前的(17.28±4.31)mmol/L、(16.36±3.92)mmol/L分别从第3、2周有显著下降(P<0.05),到第21 d时血糖分别降到(12.96±2.29)mmol/L和(11.75±2.43)mmol/L,其作用有时效性和剂量效应,而且它们的降血糖效果与药物对照组在统计学上差异无统计学意义(P>0.05)。香菇多糖能明显改善高血糖小鼠的糖耐量,其曲线下面积分别从84.58和83.34下降为68.61和65.00(P<0.05)。香菇多糖能明显增加高血糖小鼠体内的肝糖原,低、高剂量组的肝糖原水平由(22.7±3.7)mmol/L和(24.5±2.9)mmol/L提高到(28.5±3.1)mmol/L和(29.8±4.6)mmol/L(P<0.05),而且它们的作用与药物对照组相比在统计学上差异无统计学意义(P>0.05)。在实验期内香菇多糖对高血糖小鼠血清胰岛素水平的影响不明显。结论香菇多糖有显著的降血糖、改善糖耐量、增加体内肝糖原的作用,其作用是通过调节糖代谢、促进肝糖原合成、减少肝糖原分解引起的而非通过胰岛素的作用。Objective To explore the hypoglycemic effect and its mechanism of Lentinan(LTN)in alloxan-induced diabetic mice according to the therapeutics of Traditional Chinese Medicine(TCM)so as to open up a new way to treat Non-Insulin Dependent Diabetes Mellitus(NIDDM). Methods After formation of the experimental diabetic mice models by injecting alloxan with the dose of 120 mg/kg body weight,30 experimental diabetic mice(20±2g at the beginning) were divided into four groups at random,Group A were given physiological saline as control group,While Group B and C were given low dose(125 mg/kg bw)and high dose(250 mg/kg bw)LTN respectively,Group D were given Dimethyl Biguanide Hydrochloric Acid(DBHA).The effects of LTN on blood glucose concentration,glucose tolerance,the content of hepatic glycogen and serum insulin level were observed.Results LTN could reduce the blood glucose concentration significantly in alloxan-induced diabetic mice with dosage effect and time effect,and the effect of LTN was similar to the drug DBHA(Group D).LTN could also improve the glucose tolerance and increase the content of hepatic glycogen,but it could not affect the serum insulin level during the experimental period.Conclusion LTN has the curing effects to lower the blood glucose concentration,and it can improve the glucose tolerance and increase the content of hepatic glycogen in alloxan-induced diabetic mice.Its effects are not intermediary by insulin.The mechanism might be that LTN regulates the metabolism of carbohydrates by increasing hepatic glycogen synthesis or inhibiting hepatic glycogen decomposition and decreasing the absorption of glucose in the gastroenteric tract.
