摘  要::【目的】了解胃康宁对大鼠实验性胃癌的防治作用及其可能的机制。【方法】Wistar雄性大白鼠 5 0只 ,用N 甲基 N' 亚硝基 N 硝基胍 (MNNG)限期饲饮法诱导大鼠腺胃发生腺癌 ,大鼠分为三组 :阳性对照组、胃康宁 1组 (WKN 1,防癌模型 )、胃康宁 2组 (WKN 2 ,治癌模型 ) ,用光镜、电镜观察大鼠胃粘膜细胞恶性表型 ,用免疫组化法观察增殖细胞核抗原 (PC NA)、突变型 p5 3蛋白表达。【结果】胃康宁能阻止大鼠胃粘膜细胞核异形性、胞核浓染、核浆比增大等恶性表型 ,能降低PC NA和突变型p5 3蛋白表达 (与对照组比较 ,WKN 1组P <0 0 0 1,WKN 2组P <0 0 0 1)。【结论】胃康宁对大鼠实验性胃癌具有良好的防治作用 ,此作用与降低PCNA、p5 3蛋白表达有关。Objective To study the role and the possible mechanism of traditional Chinese herb WEI KANG NING (WKN) in the prevention and treatment on the experimental stomach carcinoma of rat. Methods Adenocarcinoma of adenoid stomach induced by N-methyl-N′-nitroso-N-nitrognanidine(MNNG) in 50 male Wistar rats were divided into 3 groups: positive control group, WKN 1 group(show its preventing effect) and WKN 2 group(show its therapeutic effect). The changes of malignant phenotype of rat stomach epithelial cell were observed by light microscope and electron microscope; the expression of proliferating cell nuclear antigen (PCNA) and mutant p53 protein were studied with immunohistochemistry . Results Traditional Chinese herb WKN can inhibit the malignant phenotype of stomach epithelial cell such as heteromorphism, heteropyknosis and increasing of nucleus/plasma ratio, reduce the expression of PCNA and mutant p53 protein (two groups compared with control group, both P<0 001). Conclusion WKN has a good role on the prevention and treatment on experimental rat stomach carcinoma, this role is associated with the reduction of PCNA and p53 protein.
