β胡萝卜素抗大鼠肝纤维化的实验研究

摘  要:目的 研究β胡萝卜素对四氯化碳诱导的实验性大鼠肝纤维化的治疗作用,并探讨其作用机理。方法 ①在四氯化碳诱导实验性大鼠肝纤维化3周后,同时给予β胡萝卜素25000IU/kg体重,每周两次,灌胃治疗,同时设正常及四氯化碳对照组,6周后观察肝脏的组织病理改变;②同时检测大鼠肝组织中羟脯氢酸含量;③通过免疫组织化学的方法,观察比较转化生长因子β_1(TGF_(β1))在治疗组及对照组中肝组织切片中的表达。结果 ①肝组织的普通病理切片显示,治疗组中无明显肝细胞损害,纤维化程度计分明显低于四氯化碳对照组(P<0.05);②治疗组肝组织中羟脯氨酸含量亦明显低于四氯化碳对照组(P<0.05);③免疫组织化学显示,治疗组中TGF_(β1)表达弱于四氯化碳对照组。结论 上述剂量的β胡萝卜素,能够降低由四氯化碳诱导的大鼠肝纤维化的程度,其作用途径之一可能是抑制FGF_(β1)在肝组织中的表达。Objective To study the effects and the mechanism of β carotene on rat liver fibrosis induced by carbotetrachloride(CCl4). Methods (1)After being treated with CCl4 for 3 weeks to induce the experimental rat liver fibrosis,the rats were orally feeded with β carotene 250000IU/kg body weight,twice a week,and the normal and the CCl4 control groups were set up. The liver pathological change were observed in 6 weeks.(2)The content of rat liver hydrooxylproline were observed at the same time. (3)The expression of transforming growth factorp(TGFβ1)on the slice of liver tissue were observed and compared by immunochemistry in the treated and control groups. Results (1)The pathological change was that in the treated group there was no significant liver damage and the fibrosis scores were lower than the CCl4 group(F<0. 05). (2)The content of hydrooxylproline in the treated group were decreased significantly than that in the CCl4 control group. (3) The immunochemistry showed that the expression of TGFβ1 in the treated group was weak than those in the CCl4 group. Conclusion The dosage of β carotene can reduce the severity of rat liver fibrosis significantly by inhibiting the expression of TGFβ1.
