摘  要:目的:探讨洛泊对体外培养的人胃癌细胞株SGC-7901细胞的增殖抑制作用及Akt、PTEN、ERK mRNA的表达。方法:体外培养胃癌细胞SGC-7901,用四甲基偶氮唑盐(MTT)法测定不同浓度不同时间洛铂对胃癌细胞株的生长抑制率;RT-PCR法检测Akt、PTEN、ERK mRNA表达的变化。结果:洛铂体外能抑制人胃癌细胞株SGC-7901细胞的增殖,检测发现PTEN mRNA的表达增高,AKT、ERK mRNA的表达减少,且呈剂量时间依赖性(P<0.05)。结论:胃癌SGC-7901细胞对洛泊具有药物敏感性,洛铂能诱导体外培养的胃癌细胞凋亡,其作用机制可能与洛铂诱导胃癌细胞中PTEN-mRNA的表达增高AKT、ERK mRNA的表达减少有关。Objective:To investigate the effect of lobaplatin on proliferation of gastric carcinoma cell and its relationship with the expression of AKT,PTEN and ERK mRNA.Methods:The rate of SGC-7901 cells in vitro growth inhibition was determined by MTT on different dose and time of lobaplatin.The expression of AKT,PTEN and ERK mRNA were detected with RT-PCR.Results:Lobaplatin could inhibit the proliferation of SGC-7901 cells significantly.After acting with lobaplatin,the expression of the PTEN mRNA was significantly higher and the expression of the AKT and ERK mRNA was significantly lower.They were time and concentration dependent(P<0.05). Conclusion:Lobaplatin can inhibit the growth of SGC-7901 cells and induce apoptosis,the down-regulation of AKT and ERK mRNA and up-regulation of PTEN mRNA may be one of the mechanisms of the apoptosis.
