幽门螺杆菌诱导胃上皮细胞凋亡的实验研究

摘　　要：观察幽门螺杆菌（Ｈｐ）对胃上皮细胞凋亡与增殖的影响。方法用粗制Ｈｐ总蛋白与胃上皮细胞细胞株ＭＫＮ－２８共同孵育，测定细胞增殖（ＭＴＴ染色）与细胞凋亡（ＤＮＡ梯度、流式细胞仪和Ｈｏｅｃｈｓｔ３３２５８染色法）。结果粗制Ｈｐ总蛋白能明显抑制胃上皮细胞的增殖，促进胃上皮细胞的凋亡，且此作用随着作用浓度的提高和时间的延长而增加。Purpose To observe the effect of helicobacter pylori(Hp) on the proliferation and apoptosis of gastric epithelial cell. Methods MKN 28 cell line was co cultured with soluble Hp extract for up to 72 hours,and the cell numbers(MTT assay),apoptotic cells(DNA ladder,flow cell metry,fluorchrome staining) were assessed Results The proliferation of gastric epithelial cell could be significantly inhibited by Hp,the apoptosis of gastric epithelial cell could be induced by the extract of Hp,and these effects was time and concentration dependent. Conclusions These results imply that Hp infection maybe can lead to gastric cancer and peptic ulcer through the induction of apoptosis of gastric epithelial cell.
