摘 要:目的:探讨考马斯亮兰G250法测定胃液蛋白质含量的方法和意义。方法:采用改进的Bradford法对26例正常人、8例胃癌、24例溃疡及慢性胃炎、4例肠上皮化生以及4例萎缩性胃炎患者胃液进行蛋白质含量测定。结果:G250与胃液蛋白结合后的最大吸收波长为595nm,反应体系中加入20μl10mol/LNaOH后明显提高光吸收值(P<0.05),线性范围在0~500μg/ml间,比文献报道的高,胃液蛋白质与G250结合后30min内保持稳定,本法重复性好,回收率达93.7%。结果还提示:胃癌组的胃液蛋白质含量明显高于正常对照组和其它胃疾病组(P<0.01或P<0.05)。结论:用G250法测定胃液蛋白质含量是一种可靠、简便、快速、灵敏的方法,结果具有一定的临床意义。Objective:To investigate gastric liquid protein determination using Coomassie Brilliant Blue G250 dye binding and it's clinical significance.Method:Gastric liquid protein concentration of 26 normal individuals,8 gastric carcinomas,24 gastric ulcer and chronic gastritises,4 epitheliosis and 4 atrophic gastritises were determined using modified Bradford method.Results:The max absorbance of dye reagent gastric protein complex was at 595nm;the absorbance increased after adding 20μl 10mol/L NaOH(P<0.05);linear limits of the method were 0~500μg/ml protein;the dye reagentgastric protein complex was stable within 30min,and acquired good repitition and high recovery rate(93.7%).The results also indicated that gastric liquid protein concentration of gastric carcinomas was clearly higher than that of controls and other gastric diseases(P<0.01 or P<0.05).Conclusion:Gastric liquid protein determination using Coomassie Brilliant Blue dye binding is a simple,fast and sensitive method,and it can be used in clinical diagnosis.
