摘　　要：目的：探讨考马斯亮兰Ｇ２５０法测定胃液蛋白质含量的方法和意义。方法：采用改进的Ｂｒａｄｆｏｒｄ法对２６例正常人、８例胃癌、２４例溃疡及慢性胃炎、４例肠上皮化生以及４例萎缩性胃炎患者胃液进行蛋白质含量测定。结果：Ｇ２５０与胃液蛋白结合后的最大吸收波长为５９５ｎｍ，反应体系中加入２０μｌ１０ｍｏｌ／ＬＮａＯＨ后明显提高光吸收值（Ｐ＜０．０５），线性范围在０～５００μｇ／ｍｌ间，比文献报道的高，胃液蛋白质与Ｇ２５０结合后３０ｍｉｎ内保持稳定，本法重复性好，回收率达９３．７％。结果还提示：胃癌组的胃液蛋白质含量明显高于正常对照组和其它胃疾病组（Ｐ＜０．０１或Ｐ＜０．０５）。结论：用Ｇ２５０法测定胃液蛋白质含量是一种可靠、简便、快速、灵敏的方法，结果具有一定的临床意义。Objective:To investigate gastric liquid protein determination using Coomassie Brilliant Blue G250 dye binding and it's clinical significance.Method:Gastric liquid protein concentration of 26 normal individuals,8 gastric carcinomas,24 gastric ulcer and chronic gastritises,4 epitheliosis and 4 atrophic gastritises were determined using modified Bradford method.Results:The max absorbance of dye reagent gastric protein complex was at 595nm;the absorbance increased after adding 20μl 10mol/L NaOH(P<0.05);linear limits of the method were 0～500μg/ml protein;the dye reagentgastric protein complex was stable within 30min,and acquired good repitition and high recovery rate(93.7%).The results also indicated that gastric liquid protein concentration of gastric carcinomas was clearly higher than that of controls and other gastric diseases(P<0.01 or P<0.05).Conclusion:Gastric liquid protein determination using Coomassie Brilliant Blue dye binding is a simple,fast and sensitive method,and it can be used in clinical diagnosis.
