摘 要:用放免及斑点杂交法研究缺氧对培养的冠状动脉内皮细胞(CAEC)和平滑肌细胞(CASMC)的环氧合酶(COX)和血栓素A2合成酶(TxS)基因表达的影响。实验表明在缺氧时,CAEC和CASMC中COX-1表达增加,总光密度比常氧对照组分别高3倍和1.8倍;COX-2及TXS表达无明显变化。CAEC及CASMC条件培养液中6-ketc-PGF1a含量明显升高,而TxB2与对照组无明显差异。结果提示,缺氧致冠状动脉舒张反应中可能有内皮和平滑肌细胞COX-1基因表达及PGI2生成增多的作用。Using radioimmunoassay and Dot Blot, we have studied the effects of hypoxia on the expression of COX and TxS genes and on the production of PGI2 and TXA2 in cultured coronary artery endothelialcells (CAEC)and smooth muscle cells (CASMC). The content of 6-keto-PGF1a in conditioned medium ofCAEC and CASMC exposed to hypoxia for 24 h was significantly higher than that in normoxic control group,the COX-1 mRNA levels in hypoxic CAEC and CASMC were elevated 3 and 1. 8 times respectively as high asthat in normoxic control, while the expression of COX-2 and TxS genes and the production of TXA2 in CAECand CASMC remained unchanged under hypoxia. These results suggest that the increase of COX-1 gene expression and PGI2 production in both endothelial cells and smooth muscle cells might have an effect in the mediation of hypoxic coronary vasodilation.
