摘  要:目的对直肠腺瘤miRNA表达谱进行初步研究,以期发现在直肠腺瘤发生过程中与正常直肠黏膜miRNA差异表达谱,并预测可能受其调控的靶基因。方法选择2011年1月至2011年3月于解放军第306医院肠镜检查的直肠腺瘤进行检测,筛选直肠腺瘤、正常直肠黏膜组织差异表达的miRNA,并运用生物信息软件分析其靶基因。结果通过miRNAs芯片检测,发现直肠腺瘤与正常直肠黏膜组织相比,上调的miRNA15种,下调23种。预测F3、FBXW7、HLF等可能为miR-223的靶基因。结论直肠腺瘤有其特异的miRNA表达谱,miRNA在直肠腺瘤的发生中可能起到重要作用。Objective The miRNA expression profiles in rectal adenoma are not examined till now in our country. The aim of this study is to compare and contrast the expression of miRNAs between rectal adenoma and normal tissue, and to identify some of the genes possibly associated with the development of rectal adenoma. Material and method We studied two rectal adenomas samples to examine miRNA expression profiles through miRNA microarray. We also predicted the possible targets of miRNA by bioinformatics. Result Thirty-eight miRNAs were found to be differentially expressed between rectal adenomas and normal tissues, with fifty-five miRNAs upregulated while twenty-three downregulated. Bioinformatics indicated that F3, FBXW7, and HLF might be the targets of miR-223. Conclusion There is a specific miRNA profile in rectal adenoma comparing to normal rectal tissue. miRNA pathways play an important role in the development in rectal adenoma.
