摘  要:本实验对国内首次合成的阿霉素羧甲基化葡聚糖微球(ADM─CM─DMS)进行系统的体外特性分析、人体内药代动力学特征、犬肝动脉栓塞效果的实验研究及临床初步应用。结果表明:ADM─CM─DMS具有使用方便、性能稳定、不聚块、不碎裂的特点;能缓慢释放ADM而持续有效的提高靶区药物浓度,延长局部作用时间,减少全身毒副作用;追踪观察16周犬被栓塞之肝动脉未见再通及侧枝循环形成,其栓塞作用是显著而持久的;临床初步应用治疗五例肝癌患者未发现副作用,短期追踪疗效显著。e first synthesized ADM─CM─ DMS(55~ 105um in diameter)containing ADM 30%w/w,The characteristicsof ADM─CM ─DMS in vitro and the characteristics of the phamacokinetics and embolic effect of ADM─CM─DMSin vivo were studide.The result demonstrated that ADM─CM─DMS were easier to use and did not shatter with agi-tated and permanent.ADM─CM─DMS could release ADM slowly,and prolong local target effect in liver,reducesystemic exposure to the antineoplastic agent,maintain intrahepatic drug level. Following-up angigraphic and patholog-ic results revealed that the embolic effect of ADM─CM─DMS was obvious and sustained.The blocked peripheral ves-seles did not recanalized in 16 weeks and no by-pass vesseles could be observed.5 cases with HCC were treated bytranscatheter arterial chemoembolization, no side effect could be observed,the effect of ADM─CM─DMS was obvi-ous in three months.
