白血病细胞Ki－67表达及临床意义

摘　　要：用ＡＢＣ方法检测白血病细胞Ｋｉ－６７阳性率。观察ＨＬ－６０细胞株用维甲酸前后Ｋｉ－６７阳性率的变化和各种类型的白血病患者的Ｋｉ－６７阳性率与临床预后的关系，发现加维甲酸后的白血病细胞株的总Ｋｉ－６７较加药前明显降低，并以Ｇ０／Ｇ１期降低更为明显。白血病组的Ｋｉ－６７阳性率显著高于正常对照组；急性白血病组显著高于慢性白血病组；急淋组高于急非淋组；治疗前急性白血病组中Ｋｉ－６７阳性率，缓解组稍高于未缓解组；死亡组高于缓解组。作者认为，用ＡＢＣ方法检测白血病细胞Ｋｉ－６７阳性率可以更进一步观察细胞周期中增殖细胞比例的变化，并作为白血病临床研究的一项指标。Ki-67 positive fraction in leukemic cell was measured by ABC immunostaning method,the changes of Ki- 67 positive fraction in HL- 60 cell line before and after using RA (retinoid acid) and of patients with different subtype leukemia, and the relationship between proliferating fraction and clinical prognosis were observed. We found that the total Ki-67 positive fraction of leukemic cell line after using RA for three days was strikingly lower than that before using RA. Among them, the Ki-67 positive fraction of HL-60 cell during G0/G1 phrase was evidently lower than the others. The Ki-67 positive fraction of leukemic group was strikingly higher than the control group. That of a cute leukemic group was higher than that of chronic leukemic group. That of ALL group was slightly higher than that of ANLL group. Among acute leukemic group, Ki-67 positive fraction of complete remission group was slightly higher than non-remission group. That of death group was higher than CR group. Therefore, we think that we may further observe the change of proliferating cell fraction with Ki-67 positive fraction of leukemic cell by ABC immunostaning method and use it as an index for studying leukimia clinically.
