摘 要:PHO81基因的表达受无机磷酸盐浓度的调控,属阻遏型基因。利用本组构建的PHO81-LacZ融合基因,对PHO81基因5'上游核苷酸序列作限制性内切酶缺失分析。以β-半乳糖苷酶的活力表示PHO81的表达水平,测定上游长度不同的变种表达PHO81基因的水平,提示PHO81基因起始密码上游可能存在着两个UAS功能区。The yeast acid phosphatase requlatory gene PHO 81 is a repressive gene.The expressionof the gene is in response to the presence or absence of exogenous inorganic phosphate. Tostudy its regulatory mechanism,we have sequencde 5 noncoding region 1244bp of the PHO81 gene. The coding region was fused with LacZ gene and the gene expresional level was as-sayed by measuring beta galactosidase activity. Using restriction endonuclease,the 5'-flank-ing sequences of PHO 81 were deleted. The expressional level of various deletion derivativesshowed there were two upstream activating sequences(USA)locating at -402 to -1012 and-289 to -402 upstream of the initiation codon.
