Belmont, California 94002 USA Abstract Adrenomedullin(AdM)is a newly isolated peptide with hypotensive activity in nor-motensive rat.Nothing was known regarding its role on hypertensive animals.This paper showed that AdM could reduce blood pressure in a manner of dose-depencence and its hypotensive effect on spontaneously hypertensive rat and renal hypertensive rat was more potent than that on nor- motensive rats.An antagonist for calcitonin gene related peptide(CGRP)had no influence on the effect of AdM but significantly inhibited the depressive effect of CGRP on blood pressure of hyper-tensive rat,suggesting that the hypotensive mechanism of AdM was not through the function of CGRP.AdM could markedly inhibit endothelin and angiotension Ⅱ release from aortic artery and dilate this blood vessel.The dilator effect of AdM was completely abolished by blocking the pro-duction of nitric oxide(NO)and partially decreased by inhibiting the production of prostacyclin (PGI2).Our studies suggest that (1) AdM is possibly a newly found anti-hypertensive peptide dif-ferent from CGRP;(2)its hypotensive mechanism could be related to inhibition of endothelin and angiotensin Ⅱ release and(3)its vasodilator role depends on NO and/or PGI2.
