摘 要:本研究对生长抑素(SS)在人胃癌细胞生长中的作用及机制进行探讨。将培养的人胃癌细胞MGC80及SGC7901与SS和/或hEGF(人表皮生长因子)共同孵育后,进行3H-胸腺嘧啶摄取计数和EGF放射受体分析,以观察SS对人胃癌细胞生长的作用及对EGF受体的调节作用,同时探讨SS对细胞多胺合成的影响。结果发现,SS能显著抑制两株人胃癌细胞的生长,且能抑制hEGF的促人胃癌细胞生长的作用,并首次发现,SS对EGF受体有下调作用,能使EGF受体结合率与对照组相比下降40%;我们还首次发现,SS能抑制EGF的促细胞多胺合成作用。提示SS和hEGF在人胃癌细胞生长过程中的相互影响和相互作用,以上结果表明,SS对人胃癌细胞生长的抑制作用的至少部分是由于它对EGF的负性影响,这对阐明肽类激素不同细胞内传导途径间的相互作用,具有重要的理论意义,也为肿瘤的治疗展示了某些新的前景。Recently, the growth factor-like effects or growth-inhibitory effects of some GI hormones have been noticed but their mechanisms and inter-relationship has not yet been elucidated. In this study, the effect of SS on the growth-promoting action of EGF and polyamine synthesis were examined in human gastric cancer cells. The results showed that:(1) SS obviously inhibited the growth of human gastric cancer cells either alone or in the presence of 10-8 M hEGF.(2) SS also down-regulated EGFRs on these cell lines. The specific binding of EGF decreased by 40%.(3) hEGF significantly increased the synthesis of polyamine in human gastric cancer cells(MGC80). SS partialy inhibited the effect of hEGF on polyamine synthesis. Conclusions: SS interacts with hEGF in the regulation of gastric cancer cell growth, either by down-regulating EGF receptors, or by inhibiting EGF-promoted polyamine synthesis. These findings have theoretical implications in elucidating the interactions between different intracellular signal transduction pathways.
