摘 要:用葡聚糖包裹碳饱和分析法,测定两系人乳腺癌细胞BCap-37和McF 7的雌激素受体(ER)状态,通过流式细胞仪测定其DNA含量,了解细胞增殖动力学特点,并分别用克隆形成分析法按单击多靶和线性二次方程(L-Q)模型,分析放射敏感性差异。结论是:ER阴性的乳腺癌细胞DNA含量增高,细胞组织学分化程度较差,它的放射敏感性比ER阳性乳腺癌细胞略高,该差别的主要原因是Dq(或a成份)的不同。This study is to provide the primary clue of the correlation between radiosensitivity and the estrogen receptor (ER) status. The ER assay was performed by the Dextran Coated Charcoal procedure, which shows that the McF-7 is ER positive (21. 2 fmol/mg protein), and the BCap-37 is negative 0 fmol/mg protein. The radiosensitivity of the two lines was investigated by clonogenic assay and analysed using the multi-tareget single hit model and the linear quadratic formula. The DO and Dq value of BCap-37, 138 cGy and 138 cGy, are respectively smaller than those of McF-7 , 145 cGy and 171 cGy, with difference larger in Dq than in D0. According to L-Q model, it also shows that BCap-37 is more radiosensitive than McF-7, with main disparity in the a component. Flow cytometry was used to measure the DNA index and the cell cycle kinetics. It was found that there were more aneuploid tumor cells in the BCap-37 cell line and its fraction of G2 + M phase cells is higher than that of the McF-7 cell line.
