摘 要:本研究探讨了几种特异性加压素受体拮抗剂对缺血性脑损伤的影响。结果表明,侧脑室注射 V_2,V_1/V_2受体拮抗剂可显著减轻沙土鼠的缺血性脑水肿,用 V_1受体拮抗剂则无类似效应。且中枢应用 V_2,V_1/V_2受体拮抗剂后,各脑区 cAMP 含量均显著低于缺血对照组,脑组织的 Na+-K+ATP 酶活力显著升高;而 V_1受体拮抗剂则均无明显影响。提示特异性的加压素 V_2受体拮抗剂对缺血性脑损伤起着一定的保护作用.其机理可能是通过对加压素 V_2受体的拮抗,导致受体的细胞内机理改变,产生保护脑细胞膜 Na+-K+ATP 酶活力的效应,从而发挥其对缺血性脑损伤的保护作用。This experiment attempt to c。larify the effect of several specific vasopressin receptor antagonists on ischemic brain damage.The result showed that intracerebroventricular injection (ICV) of V_2 and V_1/V_2 receptor antagonist respectively decreased significantly the ischemic brain edema of gerbils,while V_1 receptor antagonist had no such effect.And the contents of cAMP in ischemic brain regions were remarkably decreased,the Na^+-K^+ATPase activity of ischemic brain tissue was significantly increased after ICV of V_2 and V_1/V_2 receptor antagonist as compared with that of ischemic control.However,the cAMP contents and the Na^+-K^+ ATPase activity of brain tissue had no change after ICV of V_1 receptor antagonist.These suggested that the specific V_2 receptor antagonist might play a protective role in ischemic brain damage,and this role might be completed by the antagonism on V_2 receptor,then protecting Na^+-K^+ ATPase activity of brain cell membrane.
