细胞化学观察体外培养骨髓基质细胞的羟脯氨酸-2-表异构酶活性

摘  要:本实验利用液体静置法体外培养小鼠骨髓基质细胞.对生长细胞进行倒置显微镜下的原位连续观察及数种细胞化学观察.结果发现,骨髓游高细胞在两周内逐日减少,而贴壁细胞逐日增殖和分化.网状细胞的生长过程中出现很强的羟脯氨酸-2-表异构酶(Hydroxyproline-2-epimerase,EC5.1,1,a简称HP-II-E)活性.这证明网状细胞在其功能蛋白质的合成过程中有杂环亚氨基酸-羟脯氨酸参与.Murine bone marrow stromal cells were incubated in vitro by means of stationary liquid method. The cultural cells were observ d continuously in the same position under the inverted microscope at different intervals, and then a few of enzymes were shown in the developing stromal cells through enzymic cytochemistry. The result indicated that the free cells of bone marrow reduced daily in two weeks but attaching reticular cells proliferat ed and differentiated gradually under the same cultural condition. during the growing period of reticular cell, HP-Ⅱ-E(Hydroxyproline-2-epi erase, EC5, 1, 1, a) expressed very high activity. This demonstrated that Hydroxyproline i volved in the synthesis of functional proteins of reticular cells.
