野罂粟总硷抗豚鼠实验性哮喘

摘　　要：野罂粟总硷显著松弛鸡卵蛋白致敏豚鼠离体气管平滑肌,完全消除抗原攻击此标本引起的收缩反应。此作用不能被心得安所阻断。野罂粟总硷松弛支气管平滑肌的效价和效能与氨茶硷相似,清醒致敏豚鼠吸入雾化鸡卵蛋白引起发作性呼吸困难和气管伊文氏蓝渗出量增加,野罂粟总硷对此实验性哮喘具有显著的抑制作用。使发作潜伏期比对照组延长近5倍,伊文氏蓝渗出量下降56.3％。此结果表明野罂粟总硷对实验性支气管哮喘具有良好的抑制作用。此作用原理可能和激活β-肾上腺素受体及抑制抗原抗体反应引起的肥大细胞脱颗粒无关。Toral alkaloid of papaver nudicaule (TAPN)obviously reduced the muscular tension of ovalbnmin-sensitizd guinea pig trachea and abolished the contractile responses of rhe preparations to antigen challenge in vitro.These inhibitory effects were unsensitized to propranolol, The potencies and efficiencies of relaxation of trachobronchial smoofh muscle of the TAPN were similar with that of aminophylline,Inhation of the aerosol antigen induced a dyspnea attack in the concious sansitized guinea pigs and increased in Evans blue extraction in trachea of the animals in vivo.Intensity of the respira tory responses were greatly reduced in TAPN-Pretreated animals, in which TAPN prolonged the lantency time to the first bronchosbasm about 5 times and becreased the extractiom by 56.3 % compared with those of controls.These results indicate that TAPN has potent inhbitory effect on the experimental asthma, the mecha of which might be no related to activation of β—adrenoreceptors and inhibition of relebse of mast cells induced by antigen-antibody reacti on
