消化道肿瘤细胞与外周血淋巴细胞体外药敏试验相关性的研究

摘　　要：目的:通过MTT法检测外周血淋巴细胞能否替代消化道肿瘤细胞进行体外药敏试验,为不能进行肿瘤细胞药敏检测的消化道肿瘤患者提供化疗依据。方法:消化道肿瘤患者术前取血,术中取肿瘤组织,均采用单层细胞培养法对临床常用的7种抗肿瘤药进行药敏实验,以MTT法进行检测。结果:7种化疗药物体外对肿瘤细胞与外周血淋巴细胞抑制率比较,除卡铂、顺铂P<0.05,其余5种均差异无统计学意义;外周血淋巴细胞与肿瘤细胞对各化疗药药敏符合度为72.5%～85.0%,药敏相关性r值为0.506 9～0.745 8,P值均<0.01。结论:消化道肿瘤患者肿瘤细胞和外周血淋巴细胞药敏结果存在着正相关性,在无法获取肿瘤标本时,外周血淋巴细胞可替代肿瘤细胞进行药敏试验。OBJECTIVE： To study whether lymphoeytes of gastrointestinal tumor patients can take place of tumor cells to select the sensitive drugs with MTT assay,and investigate the evidence of chemotherapy strategy for the gastrointestinal tumor patients who can＇t carry out chemosensitivity of tumor cells. METHODS： Obtaining the lymphocytes before operation and tissue of tumor, sensitivity and inhibiting rates of 7 kinds of anti-cancer drugs were determined by means of primary cell culture and MTT assay. RESULTS： The effects of inhibition rate on tumor cells and lymphocytes had no significant difference between 7 chemotherapeutic agents, except cisplatin and carboplatin （ P 〈 0. 05 ） ; The compliance lymphocytes and tumor cells was 72.5%--85.0% among these of this chemotherapeutic drugs sensitivity （r was 0, 506 9--0. 745 8,P〈0. 01）. CONCLUSION： When the tumor cells of gastrointestinal tumor patients can＇t obtain to carry out chemosensitivity,the lymphocytes can take place of it.
