摘  要:目的检测兔脊髓冲击伤后早期神经细胞Bcl-2及Bax基因表达,并与原位末端标记检测的基因断裂作比较,评价两者在检测脊髓神经细胞的凋亡方面的意义和一致性。方法建立50只T9、10段脊髓损伤的兔脊髓冲击伤模型,随机分为5组(n=10),分别在伤后4、12、24、48、72 h不同时间点,分别取各组脊髓组织检测Bax、Bcl-2的表达,并与原位末端标记检测细胞凋亡进行比较,另取10只家兔作对照组。结果 Bax在伤后4 h呈阳性表达,表达高峰在伤后24~48 h,Bcl-2在伤后12 h呈阳性表达,表达高峰亦在伤后24~48 h,而但表达强度始终弱于Bax;TUNEL检测的基因断裂的变化趋势与Bax趋势相似。结论兔脊髓冲击伤后早期促进凋亡因子表达占主导,而保护性因子的表达相对不足,基因断裂,促进脊髓神经细胞凋亡。Objective To detect the expression of Bcl-2 and Bax and comparison with apoptosis detected by TUNEL in the eady stage of the spinal cord blast injury in rabbits, and evaluate the significance of both sides in spinal cord nerve cells apoptosis. Methods Fifty models of the impact of Tν9.10spinal cord injury were established, then 50 rabbits were randomly divided into five groups (n=10), and the expression of Bax, Bel-2 after 4 h, 12 h, 24 h, 48 b and 72 h was detected respectively. Apoptosis detected by TUNEL was compared. Ten normal rabbits were treated as control group. Results The expressions of protein Bax was observed 4h after in the spinal cord blast injury and reached peak value during 24-48 h. The expressions of protein Bcl-2 were observed 12 h after the spinal cordblast injury and the degree of the expressions was weaker than Bax protein, trend of DNA breaks detected by TUNEL was similar with the trend of Bax. Conclusion Because the expressions of apoptosis factor are stronger than the expression of the protective factor, and DNA breaks, apoptosis of neurons took place.
