p73 4G14C-4A14T基因多态性与肺癌发病风险相关性的系统评价

摘　　要：目的:利用Meta分析的方法,系统评价细胞周期调控基因p73 4G14C-4A14T基因多态性与肺癌易感性的关系。方法:检索2000-2010年Cochran图书馆、中国期刊网、中国生物医学文献数据库、维普科技期刊全文数据库、PubMed、Web of science、BIOSIS Pre-view、Joural Citation Reports、FMJS和EMMC数据库等,语种不限,获取有关p73 4G14C-4A14T基因多态性与肺癌发病风险相关性的研究结果,分别以病例组和对照组的p73 4G14C-4A14T基因型分布以及比值比(OR)为效应指标,确定纳入标准,对文献进行评价筛选,异质性检验,然后利用Rev Man 5.0软件对各研究原始结果进行统计处理,并计算合并的OR值及其95%可信区间(95%CI)。结果:按照纳入标准,最终入选4篇文献,累计病例2 924例,对照3 136例。Meta分析结果合并OR值为0.92,95%CI为0.69～1.22,z值为0.60。结论:说明p734G14C-4A14T基因多态性与肺癌发病风险之间无明显相关性。结果表明携带p73 4G14C-4A14T多态性不会增加肺癌的发病风险。OBJECTIVE： To evaluate relationship between polymorphism of p73 4G14C- 4A14T and susceptibility to lung cancer by means of Meta-analysis. METHODS： Studies from 2000 2010 were retrieved through Cochrane Library, Medline, Elsevier, Ovid, Highwire Press, PubMed, web of science, BIOSIS Preview, Joural Citation Reports, FMJS, EMMC database without language limitation relevant pa pets. Odds ratio of p73 4G14C -4A14T genotype distributions in the group of patients with lung cancer and the group of healthy control was calculated. The Meta-analysis was applied with RevMan 5 software for Heterogeneity test and pooled OR calculation. RESULTS： Four case-control studies with 2 924 cases and 3 136 controls were analyzed by random effect model meta analysis method. The pooled OR of p73 4G14C-4A14T genotype GC/GC vs GC/AT ＋ AT/AT was 0. 92 （95% CI： 0.69-1. 22）, and z statistics were 0. 60. CONCLUSIONS： There is enough evidence currently to prove that there is no relationship between p73 4G14C-4A14T genetic polymorphism and susceptibility to lung cancer. It is demonstrated that the genetic polymorphism of p734G14C-4A14T is not the important risk factor for susceptibility to human lung cancer.
