摘 要:目的:探讨成纤维细胞在裸鼠肺癌生长和转移过程中的作用。方法:将裸鼠40只随机分成实验组和对照组两组,将H460细胞、H460细胞+成纤维细胞分别接种裸鼠左侧胸腔,观察两组裸鼠共3周。统计其生存率,测量瘤质量,蛋白质印迹法测定动物肿瘤组织内基质金属蛋白酶1(MMP-1)的表达水平,明胶酶普法检测血清中基质金属蛋白酶-2(MMP-2)的表达。结果:裸鼠肺癌胸腔内接植成瘤率为100%,胸腔种植瘤模型逐渐出现远处转移。对照组于第18、19、20和21天共有4只裸鼠死亡,实验组分别于第12、15和19天共有3只死亡,第18、20天各有2只死亡,第21天有3只死亡,差异有统计学意义,P=0.004 8;实验组裸鼠死亡时间均较对照组早。实验组瘤质量(1.711±0.175)高于对照组(1.521±0.369),P=0.000。实验组动物血液中MMP-2的表达及瘤组织内MMP-1表达水平明显高于对照组,P<0.05。结论:成纤维细胞能够促进裸鼠肺癌的生长和转移,其作用机制可能是同肿瘤细胞相互协同,促进裸鼠体内MMP-1、MMP-2的表达有关。OBJECTIVE: To evaluate the effects of human fetal lung fibroblasts (HFL) cells on tumor growth and metastasis of lung cancer in athymic mouse. METHODS: Forty athymic mouses were randomized into experimental group and gelatinase zymography control group, H460 cells along and H460 with HFL were inoculated into the thoracic cavity of athymic mouse. After three weeks, calculated the transplantation tumor weights and mice life span, meanwhile, MMP 2 in the blood gelatinase zymography of the mou ses were detected by gelatinase zymography and MMP-1 in tumor tisues were detected by Western blot assay. RESULTS.. The rate of forming tumor was 100%, and the thoracic cavity implant models gradually appeared distance metastases. Four athymic mouse of control group died in the 18th,19th, 20th and 21st day, There were three athymic mous of experimental group died in 12th, 15th, 19th, four in 18th,20th, and three in 21st, The average survival time of the experimental group was less than that in the control group (P 0. 004 8). The transplanted tumor weights in experimental group were more than that in the control group(1. 711±0. 175 vs 1. 521± 0. 369, P=0. 000). The secretary of MMP-2 in blood and MMP-1 in tumor tisues were higher than control group(P〈0. 05). CONCLUSlON: It demonstrats that HFL cells interacting with H460 cells can increase the grouth and metastasis of lung cancer in athymic mouse, and one of the important mechanisms involves in up-regula ring the level of MMPs expression.
