摘  要:目的探讨骨癌痛大鼠脊髓背角T细胞死亡相关基因8(TDAG8)表达的变化。方法雌性未交配SD大鼠224只,体重150—180g,采用随机数字表法,将大鼠随机分为3组:正常对照组(Ⅰ组,n=64)、生理盐水组(Ⅱ组,n=64)和骨癌痛组(Ⅲ组,n=96)。采用左侧胫骨骨髓腔内接种WMker256乳腺癌肿瘤细胞的方法建立骨癌痛模型。Ⅲ组于接种前1天(基础状态)及接种后第t、3、6、9、12、15、18天、Ⅰ组和Ⅱ组于相应时点取8只大鼠,测定左后肢机械缩足阈值(MWT)。Ⅲ组于接种前1天(基础状态)及接种后第6、9、12、15、18天各处死16只,Ⅰ组和Ⅱ组相应于接种后第18天处死16只,取L4-6脊髓,免疫组化染色法测定脊髓背角TDAG8阳性细胞计数,实时,定量PCR法检测脊髓TDAG8mRNA的表达。结果与I组及基础值相比,Ⅲ组接种后第6~18天MWT降低,脊髓背角TD-AG8阳性细胞计数升高,脊髓rIDAG8mRNA表达上调(P〈0.01),Ⅱ组差异无统计学意义(P〉0.05)。结论骨癌痛大鼠脊髓TDAG8表达上调,该变化可能是骨癌痛形成的机制。Objective To investigate the changes in the expression of T-ceU death-associated gene 8(TD- AG8) in spinal cord in rats with bone cancer pain. Methods Two hundred and twenty-four female rats weighting 150-180 g were randomly divided into 3 groups: normal control group(group Ⅰ , n = 64), normal saline group (group Ⅱ , n = 64) ,bone cancer pain group(group Ⅲ , n = 96). Bone cancer pain was induced by inoculating Walker256 mammary gland carcinoma cells into the tibia medullary cavity. Mechanical withdrawl threshold(MWT) was measured at 1 d before (baseline) and 1,3,6,9,12,15 and 18 d after inoculation. Sixteen rats were sacrificed at 1 day before(baseline)and 6,9, 12, 15 and 18 d after inoculation in group m and 18 d after inoculation in groups Ⅰ and Ⅱ . The L4-6 spinal cord were removed, and the number of TDAG8 positive cell was counted, and the expression of TDAG8 mRNA was measured by RT-PCR. Results Compared with baseline value and group Ⅰ , MWT was decreased, and the number of TDAG8 positive cells and the expression of TDAG8 mRNA in spinal cord were increased at 6-18 d after inoculation in group m ( p 〈 0.01 ). Conclusion The expression of TDAG8 in spi- nal cord is up-regulated in rats with bone cancer pain, which may be involved in the mechanism of the development of bone cancer pain.
