β-连环蛋白和DNMT1在子宫内膜腺癌中表达的关系及意义

摘　　要：目的探讨β-连环蛋白(β-catenin)和DNA甲基转移酶1(DNMT1)在子宫内膜腺癌中表达的关系和意义。方法采用免疫组化方法检测β-catenin和DNMT1在86例子宫内膜腺癌中表达情况。结果在86例子宫内膜腺癌中,有54例标本(54/86,62.80%)出现β-catenin的异常表达,并与子宫内膜癌的低分化程度相关(P=0.034)。DNMT1在正常子宫内膜腺体中未见表达,而在子宫内膜腺癌中则出现了阳性表达(38/86,44.19%),并与子宫内膜腺癌的低分化程度(P=0.027)和高FIGO分期(P=0.027)相关。β-catenin的异常表达与DNMT1的表达显著相关(r=0.588,P<0.001)。结论β-catenin的异常表达与子宫内膜腺癌的低分化程度相关;DNMT1的阳性表达与子宫内膜腺癌的低分化和高FIGO分期相关;DNMT1与β-catenin的异常表达正相关。Objective To investigate the expression and correlation of β-catenin and DNA methyltransferase 1 （DNMT1） and their relationships with the clinicopathological factors of endometrial adenocarcinomas. Methods The expressions of β-catenin and DNMT1 were ex- amined in 86 cases of endometrial adenocarcinoma tissues using immunohistochemistry. Results In 86 cases of endometrial adenocareinomas,The eetopic expression of β-eatenin was observed in 54 cases （54/86,62.80%）. The ectopic expression of β-eatenin was correlated with the poor differentiation of endometrial adenoeareinomas （P = 0.034）. The expression of DNMT1 were negative in normal endometrium tissues, but positively expreseed in many eases of endometrial adenocarcinoma tissues （38/86,44.19%）. The positive expression of DNMT1 was correlated with the poor differentiation （P = 0.027 ） and high FIGO stage （P = 0.027 ） of endometrial adenocareinomas. The eetopie cytoplasmic expression of β-eatenin was significantly correlated with the positive expression of DNMT1 （r = 0.588, P 〈 0.001 ）. Conclusions The eetopic expression of β-catenin was correlated with the poor differentiation of endometrial adenocarcinoma;the positive expression of DNMT1 was correlated with the poor differentiation and high FIGO stage of endometrial adenocareinomas. The cytoplasmic accumulation of β- catenin was correlated with the expression of DNMT1.
