摘　　要：目的探讨星形细胞上调基因-1（AEG-1）在脑胶质细胞瘤中的表达及其意义。方法常规提取51例胶质瘤及13例正常脑组织中总RNA。逆转录为cDNA后，应用实时荧光定量聚合酶链反应（PCR）及免疫组织化学方法比较AEG-1基因mRNA在胶质瘤组织与正常脑组织中的表达。结果实时荧光定量PCR检测发现，在mRNA水平，AEG-1基因的表达在胶质瘤组织中明显高于正常脑组织（1．04±0．27比7．04＋-0．43，P〈0．01），并且I～Ⅱ级的胶质瘤组织中AEG-1表达的阳性率明显低于Ⅲ～Ⅳ级（4．28±0．21比8．35±0．31，P〈0．01）。免疫组织化学检测结果表明，AEG-1蛋白主要在胶质瘤瘤细胞胞质内表达，相关的表达特征与mRNA的表达一致。结论AEG-1基因mRNA和蛋白在胶质瘤组织中明显高表达，而且表达水平与肿瘤恶性程度呈正相关，其可能是胶质瘤发病过程中的恶性标志物之一。Objective To study the expression of astrocyte elevated gene-1 （AEG-1） in glioma of human brain and its significance. Methods Total RNA of 51 samples of glioma tissue and 13 samples of brain tissue of cerebral trauma after deeomp ression operation was extracted by routin Trizol way. After total RNAs were transcripted into cDNA, the mRNA expression of AEG-1 gene was detected in these samples by Real-time polymerase chain reaction （PCR）. Subsequently, The expression of AEG-1 in samples of glioma and brain tissue of cerebral trauma was examined by immunohistoehemical staining. Results Compared to normal brain tissue samples, the mRNA expression of AEG-1 was markedly increased in glioma. Immuno- histochemistry detection found that AEG-1, an intraeytoplasm expressed gene, showed higher expression in glioma eornpared with normal brain tissue （ 1.04 ± 0. 27 vs. 7.04 ± 0. 43, P 〈 0. 01 ）, and AEG-1 positive rate in I -II stage glioma was significantly lower than that in III-IV stage glioma （4. 28 ±0. 21 vs. 8. 35 ± 0. 31, P 〈0.01）, which was consistent with the characteristic of its mRNA expression. Conclusion mRNA and protein of AEG-1 were showed to be markedly high expression in samples of glioma, and associated with the progression of glioma, which suggested that AEG-1 may be one of malignant biomarkers in the pathogenesis of glioma.
