摘 要:目的建立1种测定香椿叶中芸香苷含量的反相高效液相色谱法,为其内在质量控制提供参考。方法采用Thermo ODS Hypersil column色谱柱(4.6mm×150mm,5μm),流动相为乙腈-0.1%甲酸水溶液(16∶84),流速为1.0mL/min,检测波长为254nm,柱温为25℃。结果在以上色谱条件下,供试品溶液中的芸香苷与干扰成分完全分离,其在0.0104 4~0.261 0mg/mL范围内线性关系良好(r=1),重复性实验RSD=1.77%(n=6),平均加样回收率为96.9%~98.7%(n=3),RSD为3.56%~4.42%;试验测得2个批次香椿叶中芸香苷的含量分别为(0.673±0.010)mg/g和(0.498±0.005)mg/g。结论该方法准确,快速,重复性好,可用于香椿叶中芸香苷的含量测定。OBJECTIVE To establish a reversed phase-high performance liquid chromatography(RP-HPLC) method for the quantization of rutin in Toona Sinensis Folium with the purpose of providing a reference for its quality assessment.METHODS Thermo ODS Hypersil column(4.6 mm×150 mm,5 μm) was employed for the analysis.Mobile phase was composed of acetonitrile and water containing 0.1% formic acid at the ratio of 16 to 84.Flow rate was set at 1.0 mL/min,wave length for detection was 254 nm and column temperature was 25 ℃.RESULTS Under the above chromatographic conditions,rutin has been separated from other interferences.A strong linear relationship between peak area and concentration of rutin was observed within the range of 0.010 44~0.261 0 mg/mL(r=1).The repeatability was found to be 1.77%(n=6) and recovery of rutin was from 96.9% to 98.7%(n=3).In addition,the method was also used for two batches of samples and the contents were(0.673±0.010) mg/g and(0.498±0.005) mg/g,respectively.CONCLUSION An efficient and reliable RP-HPLC-DAD method was established,which can be used for routine analysis of rutin in Toona Sinensis Folium and to assist in quality control of this herb.
