摘  要:目的探讨1,2-二甲基-3-羟基-4-吡啶酮(DFP)对AlCl3染毒小鼠肾脏三磷酸腺苷酶(ATPase)、黄嘌呤氧化酶(xanthine oxidase,XO)的影响。方法 50只健康昆明种小鼠,按体重随机分为5组:阴性对照组、铝染毒组、DFP低、中、高剂量组。小鼠经三氯化铝水溶液[200 mg/(kg·d)]灌胃4周染毒后,DFP低、中、高剂量组分别灌胃DFP水溶液[17.58,35.15,70.30 mg/(kg·d)]1周。测定小鼠肾脏铝含量和ATP酶、黄嘌呤氧化酶活性。结果三氯化铝染毒后小鼠肾脏铝含量,铝染毒组,低、中、高剂量组均高于阴性对照组(P<0.01,P<0.05);低、中、高剂量组比铝染毒组有所降低(P<0.01)。肾脏黄嘌呤氧化酶活性比较,铝染毒组高于阴性对照组(P<0.01),DFP各组均低于铝染毒组(P<0.01)。比较各组小鼠肾脏Na+K+-ATP酶及Ca2+Mg2+-ATP酶活性,铝染毒组均低于阴性对照组(P<0.01);低、中、高剂量组均高于铝染毒组(P<0.01)。结论口服DFP 1周可以促进肾脏铝排出,并能恢复ATP酶活性,降低黄嘌呤氧化酶活性。Objective To investigate the effect of deferiprone on the activities of kidney ATPase and xanthine oxidase of aluminum-induced mice.Methods Fifty healthy Kunming mice were randomly divided into 5 groups by body weight: negative control group,aluminum-induced group,deferiprone low-dose group,deferiprone medium-dose group,deferiprone high-dose group.The mice were treated by lavage with AlCl3 solution(200mg/(kg·d) AlCl3)for 4 weeks.Then the low,medium and high-dose groups were intragastrically administered with deferiprone(17.58,35.15,70.30mg/(kg·d) deferiprone,respectively)for 1 week.The content of aluminum and activities of ATPase and xanthine oxidase in kidney tissues were determined.Results The aluminum contents of aluminum-induced group and deferiprone low,medium and high-dose groups in kidney tissues were higher than negative control group(P〈0.01,P〈0.05),but the aluminum contents of three deferiprone-treated groups were lower than aluminum-induced group(P〈0.01).The activity of kidney xanthine oxidase in aluminum-induced group was higher than negative control group(P〈0.01),whereas the activities of kidney xanthine oxidase in three deferiprone-treated groups were lower than aluminum-induced group(P〈0.01).The activities of kidney Na+K+-ATPase and Ca2+Mg2+-ATPase in aluminum-induced group were lower than negative control group(P〈0.01) and three deferiprone-treated groups(P〈0.01).Conclusion Deferiprone taken orally for 1 week in aluminum-induced mice can promote the discharge of kidney Al,recover the activity of ATPase and reduce the activity of xanthine oxidase.
