摘  要:目的:观察吉非替尼(Gefitinib)对前列腺癌DU145细胞的生长抑制作用及对细胞表皮生长因子受体(EGFR)蛋白表达水平变化的影响。方法:不同浓度的Gefitinib(0~20μmol/L)分别作用于体外培养的前列腺癌DU145细胞24~120 h后,采用噻唑蓝(MTT)比色法检测细胞生长抑制率,流式细胞仪测细胞周期分布,蛋白免疫印迹(Western blot)检测细胞EGFR蛋白表达水平。结果:Gefitinib可以显著抑制DU145细胞的生长,呈时间-剂量依赖效应,并且细胞生长多停滞于G0/G1期,细胞EGFR蛋白表达分别下降了10.92%(2.5μmol/L)、43.71%(5μmol/L)、53.53%(10μmol/L)和85.98%(20μmol/L),差异有统计学意义(P<0.05)。结论:Gefitinib能显著抑制前列腺癌DU145细胞的生长,其机制可能与细胞生长在G0/G1期阻滞及EGFR蛋白表达水平下调等因素有关。Objective:To investigate the inhibitory effects of Gefitinib on the growth of DU145 cell and its' expression of epidermal growth factor receptor (EGFR). Methods: DU145 (2 × 10a / mL) was co-cultured with Gefitinib in different concentration (0 - 20μmol/ L) for 24- 120 h in vitro, and 3-(4,5-cimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) assay was used to measure the proliferation rate of DU145 ceils. Flow cytometry (FCM) was used to calculate the distribution of cell cycle, Western blotting was used to analyze the expression of protein EGFR. Results: Genifinib could effectively inhibit DU145 cells' growth in a time- and dose-dependent manner. FCM showed that Gefitinib could induce an arrest of cell cycle in G0/G1 phase. Western blotting showed that DU145 ap peared a high expression of protein EGFR. Conclusions: Gefitinib could significantly inhibit the growth of DU145 cells. The molecular mechanism of inhibitory effects may be related to the down regulation of expression of EGFR.
