摘 要:目的建立大鼠溃疡性结肠炎(ulcerative colitis,UC)细胞免疫反应性动物模型,观察环氧合酶-2(cyclooxygenase-2,COX-2)、基质金属蛋白酶-9(matrix metalloproteinase 9,MMP-9)在肠黏膜的表达。方法应用复合法(2,4-二硝基氯苯+乙酸)制备细胞免疫反应性UC大鼠模型;观察大鼠一般状态,结肠质量变化,大体形态黏膜损伤程度评分,HE染色病理、扫描电镜观察肠黏膜损伤程度,免疫组织化学染色观察COX-2、MMP-9的表达。结果模型组大鼠结肠质量,大体形态黏膜损伤程度评分较对照组显著增加(P<0.05),COX-2、MMP-9的表达水平显著升高(P<0.01)。结论复合法建立UC大鼠模型是一种较理想的UC模型,肠黏膜COX-2、MMP-9表达增加,其可能是UC的发病机制之一。Objective To establish a rat model with cellular immunoreactive ulcerative colitis ( UC ) and to investigate the expression of cyclooxygenase - 2 ( COX - 2 ) and matrix metalloproteinase 9 (MMP- 9 ) in the intestinal mucosa of rat with UC. Methods The rats model with cellular immunoreaetive UC were induced by compound method (2,4 - chlorodinitrobenzene and acetic acid). Measurement index included general state, colon weight, scoring of gross morphologic damage in acute inflammation, pathologic observation of tissues by HE staining, observation under scanning electron microscope( SEM ), COX -2 and MMP -9 expression by immunohistochemistry staining. Results Compared with those in control group, colon weight, scoring of gross morphologic damage, expressions of COX -2 and MMP -9 were markedly higher in model group. Conclusion The rat UC model induced by compound method is an ideal model. The increased expression of COX - 2 and MMP - 9 might be involved in the pathogenesis of UC.
