摘  要:目的探讨单核细胞趋化蛋白1(MCP-1)基因-2518G/A多态性及MCP-1水平与新疆哈萨克族人群脑梗死(CI)的关系及其与脑梗死病情轻重的关系。方法采用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)方法检测100例新疆哈萨克族脑梗死患者和100例正常对照者MCP-1基因-2518G/A多态性;应用酶联免疫吸附测定(ELISA)方法,检测两组血清中MCP-1水平。结果脑梗死组血清MCP-1(178.23±13.56)ng/L明显高于对照组(136.88±15.21)ng/L(t=4.385,P<0.05);脑梗死患者中、重型组的MCP-1水平明显高于轻型组(P<0.05);重型组的MCP-1水平明显高于中型组(P<0.05);脑梗死组GG基因型和G等位基因频率均显著高于对照组(χ2=7.439、7.853,P<0.05);脑梗死重型组G等位基因频率明显高于轻型组(χ2=7.627,P<0.05)。结论 MCP-1基因-2518G/A位点的G等位基因可能是新疆哈萨克族人群脑梗死发病的遗传易感基因,携带G等位基因的个体可能通过上调MCP-1表达而增加脑梗死的发病风险。Objective To investigate monocyte chemoattractant protein-1 (MCP-1) gene -2518 G/A polymorphism with serum MCP-1 level and its association with cerebral infarction(CI) of Hasake population in Chinese Xinjiang district and its association with disease severity. Methods With the technique of polymerase chain reaction restriction fragment length polymorphism (PCR-RFLP),100 cases of cerebral infarction and 100 normal controls(NC group) were detected on the -2518G/A polymorphism in MCP-1 gene. The serum level of MCP-1 in two groups was determined by enzyme-linked immunosorbent assay(ELISA). Results The level of serum MCP-1 in CI group (178.23±13.56) ng/L was significantly higher than that in NC group (136.88±15.21) ng/L( t =4. 385, P d0.05). The serum MCP-1 level of the middle and severe group of cerebral infarction was obviously higher than that of the slight group( P〈0.05) ,the serum MCP-1 level of the severe group of CI was obviously higher, than that of the middle group( P〈0.05). The frequencies of MCP-1 -2518GG genotype and G allele in CI group were significantly higher than those in NC group (x2 =7.439,7. 853, P 〈0. 05). In severe CI group,G allele was significantly higher than in the slight group (x2= 7.627, P 〈0.05). Conclusion MCP-1 gene -2518G/A G allele may be a genetic risk factor for CI of Hasake population in Chinese Xinjiang district. MCP-1 -2518G allele carrier may have up-regulate the expression of MCP-1 and hence it is a higher risk of CI.
