摘  要:目的:对大鼠大脑海马区注射凝聚态的Aβ25-35,观察Aβ25-35对大鼠空间学习、记忆能力的影响以及大鼠脑组织的病理学改变。方法:SD大鼠36只,雌雄各半,将大鼠分为正常组、生理盐水组(双侧海马区各注射5μl生理盐水)、模型组(双侧海马区各注射5μl Aβ25-35),注射后1周进行水迷宫实验,连续7 d,2次.d-1,检测大鼠空间学习、记忆能力的改变状况。模型制作30 d后对各组大鼠脑组织进行苏木素-伊红(HE)染色,在光学显微镜下观察各组脑组织的形态学变化。结果:行为学实验结果显示实验第5、6天模型组的潜伏期明显延长,穿越平台次数明显减少,与生理盐水组和正常组比较差异有统计学意义(P<0.05);病理学实验显示正常组海马神经元排列整齐、着色均匀,而模型组的海马神经元排列紊乱、细胞核固缩、不规则、胞浆浓缩。结论:大鼠双侧海马区注射Aβ25-35能够制作拟阿尔茨海默病模型。Objective:Injecting Amyloid-beta(25-35)(Aβ25-35) to the rat brain hippocampus,then observing rat model's spatial memory,working memory and spatial identify ability and the pathological changes of rat brains.Methods:A total of 36 SD rats,female and male respectively in half,were divided into normal group,saline group(bilateral hippocampus was injected with 5 μl saline) and model group(bilateral hippocampus was injected with 5 μl Aβ25-35).One week after the injection,Morris water maze experiment to begin was made for seven days,twice a day,rat model's spatial memory,working memory and spatial identify ability were deteced.After modeling 30 days,brain tissue of each group were hematoxylin-eosin(HE) staining,then the morphological changes of each group were observed through optical microscope.Results:Behavioral experiments showed that the model group's incubation period and cross platform number have significant difference with normal group and saline group(P〈0.05).Pathology results showed that hippocampal neurons in model group was disordered,the nucleus was pyknotic and irregular,the cytoplasm was concentrated.Conclusion: Bilateral hippocampus injection of Aβ25-35 can be used to establish Alzheimer's disease model successfully.
