摘 要:[目的]对球毛壳菌素A的提取分离和发酵工艺优化进行了研究,并采用HPLC进行定量分析,为建立产量高、成本低、无污染的规模化球毛壳菌发酵生产工艺打下基础。[结果]经均匀设计法优化出培养基配方中碳源和氮源最优组合为蔗糖17 g/L,淀粉6 g/L,蛋白胨0.5 g/L,最佳发酵时间为13 d。采用乙酸乙酯作提取剂,浸泡时间为3 d。分离纯化采用乙酸乙酯-石油醚(体积比1∶3)为洗脱剂,展开剂采用乙酸乙酯-石油醚-甲酸(体积比37∶63∶1)为最佳。[结论]通过对培养基配方的优化,球毛壳菌发酵产生球毛壳菌素A的发酵量可达到0.17 g/L,比目前已报道的文献值高13.3%。The optimization of fermentation process,extraction and separation of chaetoglobosin A were studied with HPLC analytical techniques to establish high yield,low cost,pollution-free scale fermentation process.[Results] The fermentation process of chaetoglobosin A was optimized by Uniform Design Method.The fermentation medium in the optimal combination of carbon and nitrogen source were determined sucrose 17 g/L,starch concentration of 6 g/L respectively,peptone concentration was 0.5 g/L and fermented for 13 d.The results showed that eluent ethyl acetate was the suitable solvent for the preparation of test samples while considering both extract ratio and fungicidal activity.Found eluant ethyl acetate,petroleum ether(1∶3),developing solvent system ethyl acetate,petroleum ether,formic acid(37∶63∶1) as the best separation conditions.[Conclusions] The ultimate production of 0.17 g/L under the optimum conditions could be reached,which was increased by 13.3% compared to 0.15 g/L under the original fermentation conditions.
