Trop2在舌鳞癌中的表达及其意义

摘　　要：目的:观察并分析Trop2在口腔舌鳞癌(oral tongue squamous cell carcinoma,OTSCC)中的表达情况及其临床意义,探讨Trop2作为OTSCC潜在治疗靶点的可能性。方法:用实时定量RT-PCR检测15例OTSCC组织及癌旁舌组织中Trop2 mRNA的表达,用前期自制的全人源抗Trop2单克隆抗体,经免疫组化检测Trop2蛋白在109例OTSCC组织及其癌旁舌组织中的表达情况t;检验分析Trop2 mRNA表达差异,卡方分析TROP2蛋白表达与临床病理特征之间关系。结果:Trop2的表达在OTSCC组织中高于癌旁舌组织;随着OTSCC分化程度的降低、肿瘤直径增大、癌细胞侵及肌层,Trop2蛋白表达也随之增高,但差异无统计学意义(P>0.05);同时可见,Trop2蛋白高表达多出现于OTSCC淋巴结转移和TNM分期晚的患者,差异有统计学意义(P<0.05);Trop2蛋白在OTSCC组织中的高表达,与患者性别、年龄无关。结论:Trop2高表达与OTSCC的发展及恶性程度有关,可作为临床预后危险因素之一,也可作为靶向治疗OTSCC的潜在靶点。Objective： To investigate the expression and clinical significance of Trop2 in oral tongue squamous cell carcinoma（OTSCC）.Methods： Real-time RT-PCR was used to characterize the expression of Trop2 mRNA in 15 OTSCC tissues.The expression of Trop2 protein in 109 tumor tissue samples including peritumoral tissue from patients with OTSCC was detected by immunohistochemisty.The comparison of clinicopathologic characteristics and the expression of Trop2 protein in these tissues were assessed by using the χ2 test.Results： RT-PCR and immunohistochemisty showed that the expression of Trop2 in OTSCC was higher than that in the tumor-adjacent non-malignant tissues.And the statistical analysis of clinicopathologic characteristics and immunohistochemistry by the χ2 test showed that the high expression of Trop2 protein in OTSCC was related to lymph node metastasis and the stage grouping with TNM classification（P= 0.035 and P = 0.015,individually）.In contrast,no statistically significant correlation was found for gender and age.Conclusion： The expression of Trop2 protein was related with the development and malignant level of OTSCC cancer.Trop2 could be used as a mark in clinical prognosis and as a potentially therapeutic target of OTSCC.
