摘 要:目的通过巢式PCR扩增18SSUrRNA基因片段确诊三日疟原虫的感染,减少三日疟的漏诊和误诊。方法采用18SSUrRNA基因片段,通过巢式PCR方法,用镜检确诊的间日疟和恶性疟滤纸血样本进行对照,检测1例可疑疟疾患者滤纸血样本。结果通过种间特异性引物的巢式PCR扩增后,间日疟及恶性疟确诊样本均扩增出相应的120bp和205bp特异性条带;可疑疟疾患者样本仅在用三日疟原虫特异性作引物时扩增出相应144bp条带的产物,证实患者感染三日疟原虫;用卵形疟原虫特异性引物扩增时,所有样品均未有条带。结论通过18SSUrRNA基因片段种间特异引物扩增,证实该患者为三日疟原虫感染。Objective In order to reduce misseddiagnosis and misdiagnosis, Plasmodium malariae cases can be identified by nested-PCR amplification of 18SSU rRNA gene fragements. Methods Compared with the blood samples of P. vivax and P. falciparum, a suspicious febrile patient was tested by nested PCR amplification of 18SSU rRNA gene fragments. Results By using the interspecific primers amplification, specific fregaments were amplified from the blood samples of P. vivax and P. falciparum with 120 bp and 205 bp respectively, and 144 bp from the suspicious febrile patient with Plasmodium malariae specific primers. All samples did not have the specific fregments of P. ovale. Conclusion Through nested-PCR amplification of 18SSU rRNA gene fragements, it was comfirmed that the patient was infected by P. malariae.
