摘  要:目的研究18F-FLT标准摄取值与非小细胞肺癌细胞株对特罗凯敏感性的关系。方法应用pcDNA3.1-snail和空pcDNA3.1转染SPCA-1细胞株并稳定培养,采用免疫组化和Western blot法,检测E-cadherin、vimentin、EGFR的表达,应用γ-counting well(SY-2021定标器)对细胞内外的放射性进行计数,计算18F-FLT的标准摄取值。采用MTT法检测特罗凯(erlotinib)对细胞株的生长抑制作用。结果扩增Snail后,SPCA-1细胞中snail表达增强,细胞变得细长,细胞融合度降低。E-cadherin表达水平实验SPCA-1细胞株高于对照SPCA-1细胞株(P<0.01),vimentin表达水平实验SPCA-1细胞株低于对照SPCA-1细胞株(P<0.01)。EGFR表达水平实验SPCA-1细胞株低于对照SPCA-1细胞株(P>0.01)。18F-FLT的标准摄取值实验SPCA-1细胞株高于对照SPCA-1细胞株(P<0.01),erlotinib对SPCA-1的抑制率实验SPCA-1细胞株低于对照SPCA-1细胞株(P<0.01)。结论细胞发生上皮细胞间质转换(epithelial mesenchymal transi-tion,EMT)时,对erlotinib的敏感性降低,18F-FLT的标准摄取值增加。通过摄取率的不同判断细胞所处的状态,并为临床筛选药物提供重要的实验依据。Objective To study the relationship between the uptake value of 18F-FLT,and the sensitivity of no-small lung cancer to erlotinib.Methods The cells were tranfered with pcDNA3.1 and pcDNA3.1-snail,then cultured steadily,the EMT cell model of non-small-cell lung cancer cell line was constructed.Morphological change and the expression change of E-cadherin,vimentin,EGFR were observed by the immunochemisry,western bloting,and detect the change of standard uptake value to 18F-FLT.Then the cells were cultured in the medium containing erlotinib,then the radioactivity of in cell and out cell was counted by γ-counting,calculating the uptake value of 18F-FLT.MTT assay was used to detect growth inhibition of erlotinib.Results After the proliferation of Snail,the SPCA-1 cells have undergone the EMT transformation,the SPCA-1 cells become long and thin,sparse,the degree of cell fusion decreased.The expression of E-cadherin decreased compared to the control group.While the expression of vimentin increased(P0.01),the expression of EGFR has on obviously change.Results of MTT indicates that the sensitivity of SPCA-1 cells with proliferation of Snail decreased correspondingly(P0.01),the uptake rate to 18F-FLT increased obviously after Snail proliferation(P0.01).Conclusion When the SPCA-1 cells undergone the EMT transformation and the uptake rate to 18F-FLT increase,the cell state can be distinguished by the different uptake rate,the experiment supplied an important evidence for clinical screening of indication of molecule targeted drugs.
