摘  要:雪卡毒素是一种存在于珊瑚鱼中的海洋毒素,极低剂量就会引起中毒。为了建立雪卡毒素的免疫分析方法,必须合成其人工抗原才能制备抗体。以雪卡毒素的结构类似物莫能菌素为半抗原,采用琥珀酸酐法将其改造成莫能菌素-琥珀酰半酯(MON-HS),再用混合酸酐法将其与大分子载体牛血清蛋白(BSA)和卵清蛋白(OVA)偶联,成功制备出了雪卡毒素人工抗原MON-HS-BSA和MON-HS-OVA。以MON-HS-BSA为免疫抗原免疫Balb/c小鼠,以MON-HS-OVA为包被抗原,用间接竞争酶联免疫吸附法(Ci-ELISA)检测到小鼠血清中产生了雪卡毒素的抗体,说明已成功合成雪卡毒素人工抗原,该抗原可用于制备雪卡毒素单克隆抗体。Ciguatoxin was a kind of marine toxin exists in coral fish. A trace residual might lead to ciguatera poisoning. In order to build an immunochromatography assay method,it was necessary to synthesize artificial antigen of ciguatoxin firstly. Monensin,an analogue of ciguatoxin,was reconstructed to succinyl monensin through the reaction between succinic anhydride and monensin,and then coupled to bovine serum albumin(BSA)and ovalbumin(OVA)by mixed acid anhydride method to synthesize the artificial antigen MON-HS-BSA and MON-HS-OVA. The antibodies against ciguatoxin were raised in serum of MON-HS-BSA immunized BalB/c mice by indirect competitive ELISA. The above results showed that the artificial antigen of ciguatoxin was synthesized successfully and it could be used to develop monoclonal antibodies against ciguatoxin in the future.
