摘 要:目的研究20(S)-人参皂苷Rh2对人结肠癌Caco-2和HT-29细胞增殖及周期的影响。方法采用荧光微孔法测定20(S)-人参皂苷Rh2对人结肠癌Caco-2、HT-29细胞增殖的影响;利用显微镜观察给药后HT-29细胞形态变化;流式细胞术分析20(S)-人参皂苷Rh2对HT-29细胞周期分布的影响。结果 20(S)-人参皂苷Rh2对Caco-2、HT-29细胞生长有抑制作用,并呈剂量依赖性;20(S)-人参皂苷Rh2作用48 h后,对HT-29、Caco-2细胞的半数抑制浓度(IC50)分别为19.68、26.79μg/mL;流式细胞分析结果显示,与对照组细胞相比,20(S)-人参皂苷Rh2能显著减少HT-29细胞的G0/G1期和G2/M期细胞比例,增加S期细胞比例。结论 20(S)-人参皂苷Rh2可抑制人结肠癌细胞增殖,主要原因是阻滞细胞处于S期。AIM To investigate the effects of 20(s)-ginsenoside-Rh2 on human colon cancer cells.METHODS Caco-2 and HT-29 human colon cancer cell lines were used.The antiproliferative effects of 20(s)-ginsenoside-Rh2 were assessed by fluorescence microporous assays.Cellular morphological changes were observed under microscopy.The cell cycle was measured by flow cytometry.RESULTS 20(s)-ginsenoside-Rh2 inhibited the proliferation of Caco-2 and HT-29 cell lines in a dose-dependent manner.Caco-2 and HT-29 cell lines were sensitive to 20(s)-ginsenoside-Rh2 with an IC50 of 26.79 and 19.68 μg/mL,respectively,in the microplate fluorimetric assay.From the result of flow cytometry analysis,20(s)-ginsenoside-Rh2,compared with the untreated cells,20(s)-ginsenoside-Rh2,caused significant reduction in number of cells at the G0/G1 and G2/M phases,and an increase in number of cells at the S phases.CONCLUSION 20(s)-ginsenoside-Rh2 could inhibit the growth by arresting human colon cancer cells in the S phase of the cell cycle.
