摘 要:目的考察大孔吸附树脂对吸水链霉菌-DY-1(Sterptomyces hygrocopious sp.DY-1)发酵产物—雷帕霉素的吸附与分离性能,并探讨采用树脂法对提取产物进行精制的工艺条件。方法采用静态吸附法考察了6种大孔吸附树脂对雷帕霉素的吸附能力及洗脱条件;以雷帕霉素吸附量、解析率为考察指标,采用薄层层析法及高效液相色谱法测定雷帕霉素的含量。结果大孔吸附树脂X-5对雷帕霉素的吸附能力最强,其最大吸附量质量分数为1.04%;6种洗脱剂中,乙醇和二氯甲烷的洗脱能力最强,当采用无水乙醇为洗脱剂时,解析率为99.21%。结论大孔吸附树脂X-5对雷帕霉素有较好的分离作用,可用于从发酵液中分离纯化雷帕霉素。Objective To investigate the separation and purification effects of macro-porous adsorption resin on rapamycin from fermentation broth of Sterptomyces hygrocopious sp. DY-1 ,and explore the technical conditions of extraction and purification of rapamycin by macroporous adsorption resins. Methods The methods of static adsorption and separation were used to investigate the effects of six kinds of macroporous adsorption resins on the adsorption and separation of rapamycin. The rapamycin adsorption capacity and recovery rate were used as evaluation criteria and the TLC and HPLC methods were used in the determination of rapamycin content. Results The results showed that X-5 was the best macroporous adsorption resin for the adsorption and separation of rapamycin. The maximum adsorbing capacity of X-5 for rapamycin was 1.04 % (w) ;the best eluting reagent was 100% (φ)ethanol and the elution rate was 99.21%. Conclusions Macroporous adsorption resin X-5 shows a better effect on adsorption and separation of rapamycin than other tested resins, which may have an potential value in the production of rapamycin.
