水通道蛋白1在肺腺癌细胞株A549中的表达及意义

摘　　要：目的探明水通道蛋白1在人肺腺癌细胞株A549中的表达情况，以探讨水通道蛋白1作为肺腺癌治疗靶位的可能性。方法培养肺腺癌细胞株A549，应用免疫细胞化学法、逆转录聚合酶链反应及Westernblot等方法检测水通道蛋白1表达。结果i种方法均证实肺腺癌细胞株A549存在水通道蛋白1的表达。结论肺腺癌细胞株A549中存在水通道蛋白1表达，结合已知水通道蛋白1的功能，深入探讨水通道蛋白1在肿瘤细胞增殖及迁移中的作用，将为肿瘤的发生、发展及转移提供新的机理。Objective To explore the expression of aquaporin 1 in human pulmonary adenocarcinoma cell line A549 and the possibility of aquaporin 1 as treatment target. Methods The expression of aquaporin 1 mRNA in cell line A549 was determined respectively by immunohistochemistry, reverse transcriptase-polymerase chain reaction and Western blot analysis. Results Aquaporin 1 was truly expressed in A549. Conclusions With known function of aquaporin 1, further investigation of aquaporin 1 function may identify new mechanisms of adenocarcinoma formation,development and metastasis.
