摘 要:目的检测五种寡糖在胃癌原发灶和淋巴结转移灶中的表达情况,探讨糖蛋白与转移的关系。方法收集伴有淋巴结转移的胃癌50例,行免疫组化和亲和组化染色。染色强度的判断采用了既考虑肿瘤的异质性又能客观地进行分析的半定量计分系统。统计学方法采用SAS系统软件,作配对t检验及相关分析。结果三种组织学类型原发灶和转移灶均有5种寡糖的表达,Slex在三种组织学类型的转移灶中的表达均显著高于原发灶(P<0.05),SA-GalNAc在印戒细胞癌和低分化腺癌中的表达明显高于原发灶。50例转移灶中,5种寡糖的表达均与原发灶呈正相关(P<0.01),其中相关系数最大的是含唾液酸的寡糖SA-GalNAc和Slex。结论原发灶表达唾液酸化的Slex和SA-GaLNAc高的癌细胞群更易发生淋巴结转移,这两种寡糖可作为判断胃癌预后的指标之一。To study the relationship the relationship between expression of sugar chains in human gastric carcinoma and lymphnode metastasis. Methods Fifty cases with gastric careinoma and lymph nide metasasis were studied by means of immunohis-tochemistry and ABC affinity histochemistry staining. The intensity of staining was scored by using semiquantitative scoring systemtha took into account the tumor heterogeneity,and statistical analysis was performed. SAS system soft ware was used for paired t-test and correlation anslysis. Results 5 types of sugar chains were expressed in three histologic types or primary carcinoma andmetastaic foci of lymph node. Sle and SA - GalNAc stainings were more intensive in metastatic foci of lymph noed than in primarycarcinoma in the three histologic types (P<0. 05). Five types of single-sugar chains in lymph ned metastasis had positive corre-lation to those in primary carcinoma, especially Sle' and SA-GalNAc. Conclusions The results indicate that the primary cancercells which express higher Sle' and SA-GalNAc are more likely to metastasize to lymph the. Both sialylated oligosaccharides maybe regarde as the prognostic indicator of gastric cancer.
