摘 要:目的:探讨GATA3在上皮性卵巢癌组织中的表达及其意义。方法:应用RT-PCR和Western Blotting检测30例上皮性卵巢癌标本和18例正常卵巢组织中GATA3 mRNA及蛋白水平表达情况,应用免疫组化检测肿瘤组织和正常卵巢组织中GATA3蛋白定位及表达情况。结果:RT-PCR提示上皮性卵巢癌组织中GATA3 mRNA表达升高,在卵巢癌组织和正常卵巢组织中的阳性表达率分别为90.0%和44.4%,差异有统计学意义(P<0.05);Western Blotting和免疫组化提示卵巢癌组织中GATA3在蛋白水平的表达明显降低,正常卵巢组织中定位在细胞核内,而卵巢癌组织中细胞核内表达不明显。结论:卵巢癌组织中GATA3在基因水平表达较正常卵巢组织上调,其翻译的蛋白表达却降低,可能是GATA3 mRNA合成的蛋白大多由核内转移而出,从而促进肿瘤的发生与发展,为进一步探讨上皮性卵巢癌的发病机理提供了实验依据。Objective:To explore the expression and significance of GATA3 in epithelial ovarian cancer tissue.Methods:RT-PCR and Western Blotting were used to detect the expression levels of GATA3 mRNA and protein in 30 cases with epithelial ovarian cancer and 18 cases with normal ovarian tissue,immunohistochemical method was used to detect the locations and expressions of GATA3 protein in epithelial ovarian cancer tissue and normal ovarian tissue.Results:The result of RT-PCR showed that the expression level of GATA3 mRNA in epithelial ovarian cancer tissue increased,the positive expression rates of GATA3 mRNA in epithelial ovarian cancer tissue and normal ovarian tissue were 90.0% and 44.4%,respectively,there was significant difference between the two groups(P0.05);the results of Western Blotting and immunohistochemical method indicated that the expression level of GATA3 protein in epithelial ovarian cancer tissue decreased significantly,GATA3 protein located in cell nucleus of normal ovarian tissue,while in epithelial ovarian cancer tissue,the expression of GATA3 protein in cell nucleus was not obvious.Conclusion:Compared with normal ovarian tissue,the expression level of GATA3 mRNA in epithelial ovarian cancer tissue increases,but the expression level of GATA3 protein decreases,which may be related to that most of GATA3 protein is transferred from cell nucleus,then the occurrence and development of epithelial ovarian cancer are promoted,the study provides experimental basis for further studies about the pathogenesis of epithelial ovarian cancer.
