Bcl-2、Bax蛋白产物在鼻咽癌中的表达、分布及意义

摘　　要：目的：探讨Ｂｃｌ－ ２ 、Ｂａｘ 蛋白产物在鼻咽癌（ＮＰＣ） 组织中的表达情况。方法：应用免疫组化技术Ｓ－ Ｐ法，对４２ 例ＮＰＣ和１０ 例正常鼻咽粘膜组织标本，进行了凋亡相关基因Ｂｃｌ－ ２ 和Ｂａｘ 蛋白产物检测。结果：（１） 在正常鼻咽粘膜中，Ｂｃｌ－ ２ 蛋白主要分布在基底层细胞，而Ｂａｘ 蛋白分布在基底层以上细胞；（２） 在ＮＰＣ 组织中，Ｂｃｌ－ ２ 和Ｂａｘ － ２ 蛋白产物表达阳性率为８５ ．７ ％ 和８３ ％ ，且Ｂｃｌ－ ２ 蛋白表达与ＢＰＣ颈淋巴结转移有关，但两种蛋白表达均与ＮＰＣ临床分期无关；（３） 在ＮＰＣ 组织中Ｂｃｌ－ ２／Ｂａｘ 表达比率存在个体差异。结论：Ｂｃｌ－ ２ 癌基因蛋白过度表达可能在ＮＰＣ发生、发展起一定作用；ＮＰＣ组织中Ｂｃｌ－ ２／Ｂａｘ 表达比率存在个体差异是否与ＮＰＣ放疗敏感性差异有关，有待进一步研究。Objective: To detect the expression and distribution of Bcl-2 and Bax proteins in nasopharyngeal carcinoma (NPC). Methods: The expression and distribution of Bcl-2 and Bax proteins were measured by the immunohistochemical method in biopsies of 10 normal nasopharyngeal epithelium (NNE) and 42 poorly differentiated NPC. Results: Bcl-2 protein was expressed in the basal cells and Bax protein in the suprabasal cell of NNE. The positive expression rates for Bcl-2 and Bax in NPC were 85.7% and 83% respectively. No correlation was found between the clinical stage and Bcl-2 or Bax expression ( P >0.05). However, the positive rates (96.5%) for Bcl-2 protein of NPC with nodal metastasis was significantly higher than that (61.5%) of NPC without nodal metastasis ( P <0.05). Furthermore, the ratios of Bcl-2 to Bax were different among different NPC samples. Conclusion: Bcl-2 protein expression may play some roles in the genesis and development of NPC. Further studies are to be conducted to find out whether different ratios of Bcl-2 to Bax in NPC are related to the radio therapy sensitivity of NPC.[
