摘 要:目的建立鬼针草总黄酮部位中芦丁的HPLC含量测定方法。方法采用高效液相色谱法,流动相为乙腈-四氢呋喃-冰乙酸-水(15.5∶5.5∶0.1∶117.5);流速1.0 ml/min;柱温30℃;检测波长360 nm。结果芦丁在75-376μg/ml的范围内线性关系良好,方程为:Y=16 262X-10 726,相关系数r=0.999 1;日内、日间精密度RSD均小于2.5%;平均回收率均为95.5%。结论该法准确、简便、重现性好,可用于鬼针草总黄酮部位中芦丁的含量控制。Objective To establish a method for determining the content of rutin in total flavonoid constituents of spanishneedles herb by high performance liquid chromatography(HPLC).Methods HPLC method was achieved on Sinochrom ODS-BP column with acetonitrile,tetrahydrofuran,acetic acid and water(15.5∶ 5.5∶ 0.1∶ 117.5)as mobile phase at a flow rate of 1.0 ml/min.Column temperature was kept at 30 ℃ and UV detection wave length was set at 360 nm.Results There was good linearity between the peak areas and the compound concentrations for rutin in the range of 75-376 μg/ml.The equation was Y=16 262X-10 726 with r=0.999 1.The inter-day and intra-day precision was both less than 2.5%.Mean recovery was 95.5%.Conclusion The method is simple,accurate and repeatable,and could meet the requirement of quality control of rutin in total flavonoid constituents of spanishneedles herb.
