中药复方“松友饮”增强IFN-α抑制肝癌的实验研究

摘　　要：目的:研究联用中药"松友饮"和干扰素α(IFN-α)对高转移人肝癌裸鼠原位模型肿瘤生长、转移和生存期的影响。方法:以人高转移MHCC97H细胞建立裸鼠原位移植瘤模型,48只成模裸鼠随机分成对照组、"松友饮"组、IFN-α组及"松友饮"+IFN-α组,肝脏接种肿瘤24h开始每天1次灌胃给药及皮下注射,连续5周。第35天每组随机处死6只裸鼠,计算肿瘤体积及转移情况;剩余成瘤裸鼠用于观察生存期。结果:"松友饮"组、IFN-α组及"松友饮"+IFN-α组肿瘤体积、肺转移结节数及生存期与对照组比较,均有显著差异(P<0.05)。结论:连续5周联用"松友饮"显著增强IFNα对HCC裸鼠模型肿瘤生长、转移的抑制作用并延长生存期,对HCC病人有潜在的应用价值。Objective： To investigate the combined therapy effects of Chinese herbal compound Songyou Yin and interferon–alfa（IFN-α） on tumor growth,metastasis and survival in nude mice bearing human hepatocellular carcinoma（HCC） xenografts with high metastatic potential.Methods： Metastatic human HCC nude mice models were established by orthotopic implantation with a high metastatic potential human HCC cell line MHCC97H.Forty-eight nude mice bearing orthotopic xenografts were randomized into control group,Songyou Yin group,IFN-α group and the combined group（Songyou Yin ＋ IFN-α）.Daily intragastric adminstration of Songyou Yin and injection of IFN-α were given for 5 consecutive weeks,24 hours after orthotopic implantation.Mice were sacrificed on day 35,tumor volume and metastasis were examined.The remaining mice were used to observe life span.SAS 8.2 software was applied for statistical analysis,and the significance was defined as P0.05.Results： The HCC tumor volume in Songyou Yin group,IFN-α group and the combined group（Songyou Yin＋ IFN-α）,accorded with the number of pulmonary metastatic nodules,life span had significant difference with the control group.Statistical significance could be seen as comparison was made between the combined group and the single drug therapy groups（P0.05）.Conclusion： Five weeks administration of Chinese herbal compound Songyou Yin reinforced the inhibition effects of IFN-α on tumor growth and lung metastasis,and extension of life span of nude mice bearing human HCC MHCC97H,which indicated its potential use in patients with HCC.
