摘 要:目的 探讨醒脑静注射液(XNJI)对脑缺血灌注损伤(CIRI)的防治作用及其机理。方法 制备家兔CIRI模型,随机分为对照组和XNJI组,动态观察血浆及脑组织一氧化氮(NO)水平、内皮素(ET)含量、丙二醛(MDA)浓度、超氧化物歧化酶(SOD)活性及脑超微结构的变化。结果 脑缺血再灌注期间,血浆和脑组织NO水平及SOD活性明显下降(P<005和P<001),ET及MDA含量显著升高(P<005和P<001),超微结构发生异常改变;使用XNJI后,上述各指标的异常变化明显减轻,与对照组相比有显著性差异(P<005和P<001)。结论 XNJI对CIRI具有良好的防护作用,其机制与提高机体NO水平、降低ET及氧自由基水平有关。Objective To investigate the protective effect of xingnaojing injection (XNJI) on brain and its mechanism during cerebral ischemia and reperfusion injury (CIRI).Methods Using a CIRI model in 20 rabbits which were randomly into control group and XNJI grorp,changes of several parameters were dynamically observed in plasma and brain during CIRI,which included nitric oxide (NO)、endothelin(ET)、malondialdehyde(MDA)、superoxide dismutase (SOD)and ultrastructural of brain.Results NO level and SOD activity in plasma or cerebral tissue all decreased significantly ( P <0 05 and P <0 01),ET level and MDA content in plasma or cerebral tissue increased remarkably ( P <0 05 and P <0 01),and there were obviously abnormal changes of brain ultrastructrually during CIRI;XNJI administered at 20 minutes before ischemia and at begining of reperfusion,the abnormal changes of indexes as above were ameliorated markedly,and there was significant difference between control group and XNJI grorp ( P <0 05 and P <0 01).Conclusion XNJI could effectively protect the brain from ischemia-reperfusion injury,the mechanism was related to its raising NO level and reducing ET or oxygen free radicals.
