摘 要:目的检测胰腺癌(PC)细胞株中两种代谢相关基因的单核苷酸多态性(SNPs),为临床预测氟尿嘧啶类药物的疗效及毒副反应提供参考依据。方法培养PANC-1、CFPAC、SW1990三株PC细胞,采用QIAamp DNABlood Minikit试剂盒提取基因组DNA,用基质辅助激光解吸电离飞行时间质谱技术行二氢嘧啶脱氢酶基因(DPYD)rs1801159、rs1801160、rs17376848,亚甲基四氢叶酸还原酶(MTHFR)基因rs2274976、rs1801131、rs1801133的SNPs位点检测。结果三种PC细胞株的DPYD基因rs1801159位点基因型均为A/A纯合子,rs1801160位点基因型均为G/G。CFPAC细胞株DPYD基因的rs17376848位点基因型为A/G,PANC-1、SW1990细胞株的rs17376848位点基因型为A/A纯合子。PANC-1、CFPAC细胞MTHFR基因的rs1801131位点基因型为A/C杂合子,rs1801133位点基因型为C/C;SW1990细胞株MTHFR基因的rs1801131位点基因型为A/A纯合子,rs1801133位点基因型为T/T纯合子;三株细胞MTHFR基因的rs2274976基因型位点均为G/G纯合子。结论本实验所测三株PC细胞的DPYD基因rs1801159、rs1801160位点、MTHFR基因rs2274976位点基因型相同,其余各位点基因型均不完全相同。Objective To detect single nucleotide polymorphisms(SNPs) of two pharmacokinetics related genes in three pancreatic cancer cell lines in order to provide references for prediction clinical efficacy and toxicity of fluorouracil.Methods PANC-1,CFPAC and SW1990 cell lines were cultured.The genomic DNA of the two cell lines were isolated by QIAamp DNA Blood Minikit.SNP genotyping of DPYD gene rs1801159,rs1801160,rs17376848 and MTHFR gene rs1801131,rs1801133,rs2274976 was performed by means of matrix assisted laser desorption ionisation-time of flight mass spectrometry method(MALDI-TOFMS).Results The genotypes of DPYD gene locus rs1801159 and rs1801160 were A/A and G/G respectively in the 3 cell lines.The genotype of DPYD gene locus rs17376848 was A/G in CFPAC cell line,While that were A/A in PANC-1 and SW1990 cell lines.The genotypes of MTHFR gene locus rs1801131 were A/C in PANC-1 and CFPAC cell lines,While that was A/A in SW1990 cell line.The genotypes of MTHFR gene locus rs1801133 were C/C in PANC-1 and CFPAC cell lines,While that was T/T in SW1990 cell line.The genotype of MTHFR gene locus rs2274976 was G/G in the above 3 cell lines.Conclusion Loci rs1801159,rs1801160 of DPYD gene and locus rs2274976 of MTHFR gene were same in the detected 3 cell lines.While other detected loci of DPYD and MTHFR gene in the 3 cell lines were expressed differently.
