摘  要:目的探讨鱼藤素对膀胱癌T-24细胞的抑制作用及其机制。方法以不同浓度的鱼藤素作用于人膀胱癌T-24细胞,用倒置显微镜观察细胞形态学的改变,MTT法检测T-24细胞的存活率,流式细胞术(FCM)观察细胞周期的改变,划痕实验检测鱼藤素对细胞迁移能力的影响。结果倒置显微镜观察鱼藤素处理后的T-24细胞呈现缩小、皱缩、空泡、脱壁等增殖变慢的形态学改变;MTT法显示鱼藤素显著抑制T-24细胞增殖,其抑制率呈时间及浓度依赖性;FCM检测发现鱼藤素使T-24细胞的G0/G1期延长,S期缩短,周期主要阻滞在G0/G1期;划痕实验结果显示,鱼藤素可降低T-24细胞的迁移能力。结论鱼藤素能够使T-24细胞的生长阻滞在G0/G1期,从而显著抑制T-24细胞的增殖及降低细胞的迁移能力。Objective To investigate the effects and mechanism of deguelin on human bladder cancer T - 24 cells proliferation. Methods T - 24 cells were treated with deguelin with concentrations of 0 - 1 000 nmol/L. Cell proliferation, cell cycle and migration ability of T - 24 were assessed by MTT assay, flow cytometry (FCM) and wound healing test, respectively. Results Hindered proliferation presentations, including shrink, vacuoles and deadherence, were observed in T- 24 treated with deguelin under inverted microscope. Deguelin significantly inhibited the proliferation of T- 24 following concentration and time dependent manners, according to MTT. In FCM, deguelin lengthened the G0/G1 phase and shorten the S phase in T - 24 cells. The migration capacity was receded in deguelin - treated cells compared with that in control cells. Conclusion Deguelin blocks T -24 cells at G0/G1 phase, thus significantly inhibits the proliferation and migration of T - 24 ceils.
