摘  要:目的研究大鼠创伤性脑水肿皮质中AQP-4与MMP-9的表达关系及甲强龙对其表达的影响。方法采用成年雄性SD大鼠随机分为甲强龙组(n=25)、模型组(n=25)、假手术组(n=5);甲强龙组和模型组均采用改进Feeney法制作脑损伤模型。甲强龙组致伤后即刻腹腔内给药,模型组则注射对应生理盐水。根据处死时间分为6h、24h、3d、5d、7d,5组。过度麻醉并断头取脑,对损伤皮层采用HE染色及免疫组化进行AQP4与MMP-9的检测。结果在模型组与甲强龙组中6h已可见AQP4与MMP-9的表达,MMP-9与AQP-4的表达均在3d达到高峰,模型组AQP4及MMP-9的表达显著高于甲强龙组(P<0.05),相关系数(r=0.753,P<0.01)。结论创伤性脑损伤后大鼠AQP4与MMP9的表达呈正相关;甲强龙冲击治疗能够通过下调AQP4及MMP9的表达来发挥减轻脑水肿的作用。Objective To study the relationship between the expressions of MMP-9 and AQP-4 and to explore the mechanisms of influenced by Solu-Medrol in traumatic brain edema.Methods Fifty-five healthy adult male SD rats were randomly divided into sham operation group(n=5),model group(n=25) and the Solu-Medrol treatment group(n=25).The traumatic brain injury of the latter two groups were induced by Feeney's.Solu-Medrol treatment group underwent abdomen injection of Solu-Medrol after the injury.Model group underwent abdomen injection of NS of the same amount.The rats in the model group and the MP group were divided into 5 subgroups(6h,24h,3d,5d,7d groups) of 5 rats each respectively according to different execute time.Under 10% cholralhydrate anesthesia,the group of model group and MP treatment group was killed by decapitation.The histology was detected by HE staining,the expression of MMP-9 and AQP4 was detected by immunohistochemical.Results The expression of MMP-9 and AQP4 in the Model group and Solu-Medrol treatment group appeared 6 hours after injury and their expression reached peak on day3.The expression was significantly lower in Solu-Medrol treatment group than that in model group(P〈0.05).There was a significantly positive correlation between AQP-4 expression and expression of MMP-9(r=0.753,P〈0.01).Conclusion The expression of MMP-9 correlated with AQP4 expression positively.High-dose of Solu-Medrol to treat traumatic brain edema in rats probably by inhibiting the activities of MMP-9 and AQP4.
