摘  要:目的:探讨细胞周期蛋白cyclin D1外显子4(870 A/G)多态性与慢性髓细胞白血病的相关性。方法:采用聚合酶链反应(PCR)和PCR-RFLP的方法,分析了40例慢性髓细胞白血病患者和46例健康对照者的cyclin D1基因型。结果:与健康对照组相比,携带GG基因型患者慢性髓细胞白血病的相对风险度增加3.06倍(OR=3.06,95%CI=1.033~9.043,P=0.041);按年龄分层,>45岁携带cyclin D1 GG基因型的患者相对风险度增加5.00倍(OR=5.00,95%CI=1.166~21.413,P=0.022)。结论:Cyclin D1基因多态性与慢性髓细胞白血病的发生发展有着密切相关。Objective Cyclin D1 is a key protein involved in cell cycle regulation.The cyclin D1 is located at 11q13 and is often altered in human cancers.To our knowledge,no report has been issued concerning its relationship with chronic myelocytic leukemia in China.Method The A870G cyclin D1 polymorphism was analyzed by polymerase chain reaction(PCR) and PCR-restriction fragment length polymorphism(PCR-RFLP) in 40 cases and 46 controls.Results The GG genotype of Cyclin D1 was associated with a 3.06-fold higher risk for the development of chronic myelocytic leukemia〔OR = 3.06,95% CI = 1.033 ~ 9.043,P = 0.041〕;Stratification of patients according to age showed that the GG genotype was significantly observed in older than 45 years,and chronic myelocytic leukemia with high risk〔OR = 5.00,95% CI = 1.166 ~ 21.413,P = 0.022〕.Conclusion The Cyclin D1 genetic polymorphism might be related to the occurrence of chronic myelocytic leukemia in Chinese population.
